ST:STUDY_SUMMARY                 	remains high. The metabolic state of cells that remain after endocrine therapy,
ST:STUDY_SUMMARY                 	specifically oxidative stress, is poorly understood. We demonstrate that
ST:STUDY_SUMMARY                 	endocrine-tolerant persister cells are oxidatively stressed after initiation of
ST:STUDY_SUMMARY                 	endocrine therapy. Furthermore, modulation of redox levels similarly modifies
ST:STUDY_SUMMARY                 	persister cell fitness. We find ferroptosis sensitivity and lipid peroxidation
ST:STUDY_SUMMARY                 	to be a dominant factor in the oxidative state, and persister cells are found to
ST:STUDY_SUMMARY                 	be sensitive to ferroptosis induction compared to parental cells. Persister
ST:STUDY_SUMMARY                 	cells exhibited an altered lipidome, with an increase in phospholipids having a
ST:STUDY_SUMMARY                 	predilection for peroxidation. We hypothesized that ferroptosis induction would
ST:STUDY_SUMMARY                 	be well-coordinated with endocrine therapy. Xenografts treated with a
ST:STUDY_SUMMARY                 	combination of fulvestrant and ferroptosis induction exhibited the greatest
ST:STUDY_SUMMARY                 	treatment effect, supporting the role of ferroptosis as a therapeutic strategy
ST:STUDY_SUMMARY                 	in ER+ breast cancer.
