Sampleprep Summary: | Aliquots of 200 µL human urine were mixed with 20uL of urease suspension placed on Thermomixer for 1 hour at 37°C on 1400rpm. Then added 1.7mL of methanol, vortexed for 5 minutes, centrifuged for 10 minutes on 14,000 rpm at 4°C. 1.5 mL of the supernatant was transferred to new individual 2 mL Eppendorf tubes and dried completely under nitrogen flow. Placed on vacuum centrifuge for 30 min to ensure complete dryness. Formation of methoximes by adding 10µL of 40mg/mL MeOx in pyridine to each sample. Placed onto Thermomixer for 90 min at 30⁰C. Then FAME retention index markers and MSTFA added to each sample for derivatization step. Samples returned to Thermomixer for 45 min at 70⁰C. |
