PR:PROJECT_TITLE                 	Metabolomic analysis of oxytocin effects on social deficits in mice
PR:PROJECT_SUMMARY               	The goal of this study was to determine the effects of the neuropeptide oxytocin
PR:PROJECT_SUMMARY               	(OT) on metabolomic profiles, using a mouse model of autism-like behavior, the
PR:PROJECT_SUMMARY               	BALB/cByJ inbred strain. We have previously reported that subchronic treatment
PR:PROJECT_SUMMARY               	with OT can lead to persistent reversal of social deficits in BALB/cByJ and
PR:PROJECT_SUMMARY               	other models of autism spectrum disorder (ASD). In this study, mice were given a
PR:PROJECT_SUMMARY               	subchronic regimen with either vehicle or OT, and then evaluated for social
PR:PROJECT_SUMMARY               	approach. At the end of the study, brain and blood were collected for
PR:PROJECT_SUMMARY               	metabolomic analysis. In addition, fecal samples were taken at different time
PR:PROJECT_SUMMARY               	points during the treatment and testing regimen. The results from this project
PR:PROJECT_SUMMARY               	could elucidate mechanisms underlying the prosocial effects of oxytocin, and
PR:PROJECT_SUMMARY               	identify new targets for the development of highly specific oxytocin-related
PR:PROJECT_SUMMARY               	drugs.

CO:COLLECTION_SUMMARY            	Fecal samples were collected at 4 time points: (1) 24 hr before the first
CO:COLLECTION_SUMMARY            	injection, (2) 24 hr after the first injection, (3) 24 hr after the third
CO:COLLECTION_SUMMARY            	injection, and (4) 24 hr after the 3-chamber test. To collect the samples,
CO:COLLECTION_SUMMARY            	individual mice were placed into a clean mouse cage without bedding, and one or
CO:COLLECTION_SUMMARY            	two pellets were collected per mouse. Pellets were immediately frozen on dry ice
CO:COLLECTION_SUMMARY            	and stored at -80o C. Samples were coded by treatment: vehicle-treated mice
CO:COLLECTION_SUMMARY            	(n=8) and OT-treated mice (n=12).
#TREATMENT
TR:TREATMENT_SUMMARY             	BALB/cByJ male mice (4-5 weeks in age; Jackson Laboratory, Bar Harbor, ME) were
TR:TREATMENT_SUMMARY             	given four treatments of either vehicle or OT (1.0 mg/kg; IP) across 8 days,
TR:TREATMENT_SUMMARY             	with at least 48 hours between each injection. Mice were evaluated in a
TR:TREATMENT_SUMMARY             	3-chamber choice test for social preference 24 hr after the final injection.
#SAMPLEPREP
SP:SAMPLEPREP_SUMMARY            	Fecal pellets were transferred into a pre-labeled MagnaLyser tube and massed.
SP:SAMPLEPREP_SUMMARY            	Phosphate buffer was added depending on the mass of each sample (500 uL for up
SP:SAMPLEPREP_SUMMARY            	to 100 mg and 1000 uL for more than 200 mg). Samples were homogenized using
SP:SAMPLEPREP_SUMMARY            	MagnaLyser for two 30 sec pulses and then centrifuged. The supernatant of each
SP:SAMPLEPREP_SUMMARY            	sample was transferred into pre-labeled 0.2 um nylon filter tube and
SP:SAMPLEPREP_SUMMARY            	centrifuged. A volume of each filtrate required to analyze 25 mg of sample was
SP:SAMPLEPREP_SUMMARY            	aliquoted, mixed with a volume of phosphate buffer to bring the total volume to
SP:SAMPLEPREP_SUMMARY            	630 uL, and 70 uL of Chenomx ISTD. For a subset of samples with excess material,
SP:SAMPLEPREP_SUMMARY            	a total study pool was created by mixing the remaining volume. Five replicates
SP:SAMPLEPREP_SUMMARY            	were created, and these prepared identically to the study samples. The tubes
SP:SAMPLEPREP_SUMMARY            	were vortexed for 2 min on a multi-tube vortexer and centrifuged at 16,000 rcf
SP:SAMPLEPREP_SUMMARY            	for 4 min. A 600 µl aliquot of the supernatant was transferred into pre-labeled
SP:SAMPLEPREP_SUMMARY            	5 mm (4”) NMR tubes for data acquisition on a 700 MHz spectrometer.
