Here we show ST:STUDY_SUMMARY that AMP-activated protein kinase (AMPK), a metabolic checkpoint kinase, confers ST:STUDY_SUMMARY metabolic stress resistance to leukemia-initiating cells (LICs) and promotes ST:STUDY_SUMMARY leukemogenesis. Upon dietary restriction, MLL-AF9-induced murine acute myeloid ST:STUDY_SUMMARY leukemia (AML) activated AMPK and maintained leukemogenic potential. AMPK ST:STUDY_SUMMARY deletion significantly delayed leukemogenesis and depleted LICs by reducing the ST:STUDY_SUMMARY expression of glucose transporter 1 (Glut1), compromising glucose flux, and ST:STUDY_SUMMARY increasing oxidative stress and DNA damage. LICs were particularly dependent on ST:STUDY_SUMMARY AMPK to suppress oxidative stress in the hypoglycemic bone marrow environment. ST:STUDY_SUMMARY Strikingly, AMPK inhibition synergized with physiological metabolic stress ST:STUDY_SUMMARY caused by dietary restriction and profoundly suppressed leukemogenesis. Our ST:STUDY_SUMMARY results indicate that AMPK protects LICs from metabolic stress and that ST:STUDY_SUMMARY combining AMPK inhibition with physiological metabolic stress potently ST:STUDY_SUMMARY suppresses AML by inducing oxidative stress and DNA damage.
