Specifically, we recently showed PR:PROJECT_SUMMARY both compound (rapamycin) treatment and genetic manipulation using a mechanistic PR:PROJECT_SUMMARY Target of Rapamycin (mTOR) haploinsufficient (mTOR+/-) mutant confers cardiac PR:PROJECT_SUMMARY protection against cardiomyopathies of two different etiologies in zebrafish. PR:PROJECT_SUMMARY Mechanistically, antihypertrophy, anti-apoptosis and proautophagy of mTOR PR:PROJECT_SUMMARY inhibition play important roles in this cardioprotection process. Our central PR:PROJECT_SUMMARY hypothesis is that this mTOR inhibition-mediated cardioprotection is conferred PR:PROJECT_SUMMARY by transcription factor EB (TFEB)-regulated autophagy activation. To test this PR:PROJECT_SUMMARY hypothesis, we propose to pursue a systematic metabolite profiling analysis in PR:PROJECT_SUMMARY the heart upon DOX stress under fine-tuned mTOR inhibition. We expect to define PR:PROJECT_SUMMARY metabolite signatures in different types of cardiomyopathy, metabolite signature PR:PROJECT_SUMMARY for the cardioprotective effects of mTOR inhibition, as well as metabolite PR:PROJECT_SUMMARY signatures for TFEB genetic manipulations.
