A total of 20 control and 20 CO:COLLECTION_SUMMARY glaucomatous AH samples were included for these studies. All AH samples were CO:COLLECTION_SUMMARY immediately stored at −80°C until time of use. The mean age of donors was CO:COLLECTION_SUMMARY 69.5 ± 9.9 years and both genders were included for these studies. CO:SAMPLE_TYPE Eye tissue #TREATMENT TR:TREATMENT_SUMMARY No treatment was performed on the eyes. #SAMPLEPREP SP:SAMPLEPREP_SUMMARY Aqueous humor samples were subjected to extraction of lipids using suitable and SP:SAMPLEPREP_SUMMARY minimal modification of Bligh and Dyer method. The lower organic phase SP:SAMPLEPREP_SUMMARY containing the extracted lipids was isolated and solvent dried with a Speed-Vac SP:SAMPLEPREP_SUMMARY (Model 7810014; Labconco, Kansas City, MO). Samples were subsequently flushed SP:SAMPLEPREP_SUMMARY with argon gas to prevent oxidation. Proteins recovered from the corresponding SP:SAMPLEPREP_SUMMARY upper aqueous phase were quantified using the Bradford method. A subset of SP:SAMPLEPREP_SUMMARY protein samples were also subjected to densitometric quantification using bovine SP:SAMPLEPREP_SUMMARY serum albumin (BSA) as a standard (amino acid quantified) after electrophoretic SP:SAMPLEPREP_SUMMARY separation on a PHAST (GE Healthcare Bio-Sciences AB, Sweden) gel system. We SP:SAMPLEPREP_SUMMARY repeated protein estimations using an amino acid analyzer after overnight SP:SAMPLEPREP_SUMMARY digestion in hydrochloric acid following previously published protocols. The SP:SAMPLEPREP_SUMMARY protein amounts determined using amino acid analyzer were utilized in SP:SAMPLEPREP_SUMMARY normalization of lipids per amount of proteins. In order to determine and ensure SP:SAMPLEPREP_SUMMARY extraction efficiency, ovine wool cholesterol (molecular mass 386.7; catalog no. SP:SAMPLEPREP_SUMMARY 700000; Avanti Polar Lipids, Albaster, AL) was premixed with AH prior to SP:SAMPLEPREP_SUMMARY extraction. All extractions and subsequent handling were made using glass vials SP:SAMPLEPREP_SUMMARY to avoid contaminating impurities.
