In mice, the gut microbiome contributes to PR:PROJECT_SUMMARY ozone-induced airway hyperresponsiveness, a defining feature of asthma. The PR:PROJECT_SUMMARY purpose of this study was to identify metabolites that could be mediating this PR:PROJECT_SUMMARY role. Gut bacterial enzymes modify ingested substances producing metabolites PR:PROJECT_SUMMARY that enter the blood and circulate to host tissues where they may exert a PR:PROJECT_SUMMARY variety of effects. Therefore, we performed global metabolomic profiling on PR:PROJECT_SUMMARY serum of mice after acute ozone exposure. To identify the role of the microbiome PR:PROJECT_SUMMARY in mediating ozone-induced metabolomic changes, mice were treated for two weeks PR:PROJECT_SUMMARY with a cocktail of antibiotics in the drinking water or with control water and PR:PROJECT_SUMMARY then exposed to air or ozone (2 ppm for 3 hours). Twenty four hours later, blood PR:PROJECT_SUMMARY was harvested and serum analyzed via liquid-chromatography or gas-chromatography PR:PROJECT_SUMMARY coupled to mass spectrometry. We observed marked effects of both ozone exposure PR:PROJECT_SUMMARY and antibiotics on the serum metabolome. One fraction was used for analysis by UPLC-MS/MS SP:SAMPLEPREP_SUMMARY with positive ion mode electrospray ionization.
