Pregnant women were ST:STUDY_SUMMARY recruited at two obstetrical clinics in Urbana Champaign IL from 2014-2021. ST:STUDY_SUMMARY Women were enrolled between 8-14 weeks of pregnancy. Eligibility criteria ST:STUDY_SUMMARY included 18-40 years of age, English speaking, not in a high risk pregnancy, ST:STUDY_SUMMARY planning to stay in the area until their child's first birth day and living ST:STUDY_SUMMARY within 30 minutes of the University of Illinois campus. Health, demographic and ST:STUDY_SUMMARY life style data and biospecimens were collected at 5 time points across ST:STUDY_SUMMARY pregnancy and children were followed prospectively from birth including ST:STUDY_SUMMARY assessments at birth, 1-5 weeks 4.5 months, 7.5 months, 2 years, 3 years, 4 ST:STUDY_SUMMARY years and 7.5 years of age. Physical and neurobehavioral development were ST:STUDY_SUMMARY assessed, with a particular focus on the impacts of prenatal chemical exposures ST:STUDY_SUMMARY (phthalates, phenols, PFAS) or prenatal maternal stress on child health ST:STUDY_SUMMARY outcomes. Pregnant women were CO:COLLECTION_SUMMARY recruited at two obstetrical clinics in Urbana Champaign IL from 2014-2021. CO:COLLECTION_SUMMARY Women were enrolled between 8-14 weeks of pregnancy. Eligibility criteria CO:COLLECTION_SUMMARY included 18-40 years of age, English speaking, not in a high risk pregnancy, CO:COLLECTION_SUMMARY planning to stay in the area until their child's first birth day and living CO:COLLECTION_SUMMARY within 30 minutes of the University of Illinois campus. Health, demographic and CO:COLLECTION_SUMMARY life style data and biospecimens were collected at 5 time points across CO:COLLECTION_SUMMARY pregnancy and children were followed prospectively from birth including CO:COLLECTION_SUMMARY assessments at birth, 1-5 weeks 4.5 months, 7.5 months, 2 years, 3 years, 4 CO:COLLECTION_SUMMARY years and 7.5 years of age. Physical and neurobehavioral development were CO:COLLECTION_SUMMARY assessed, with a particular focus on the impacts of prenatal chemical exposures CO:COLLECTION_SUMMARY (phthalates, phenols, PFAS) or prenatal maternal stress on child health CO:COLLECTION_SUMMARY outcomes. HHEAR Urine Pool SP:SAMPLEPREP_SUMMARY 1 (50 µL each), HHEAR Urine Pool 2 (50 µL each), CHEAR reference urine (50 µL SP:SAMPLEPREP_SUMMARY each) and NIST urine (SRM 3672) reference material (50 µL each) were provided SP:SAMPLEPREP_SUMMARY by the NC HHEAR Hub. LC-MS grade water (50 µL) was used as blanks. All samples SP:SAMPLEPREP_SUMMARY were thawed at 4°C overnight before the preparation. Samples, including study SP:SAMPLEPREP_SUMMARY samples, study pool samples, HHEAR reference urine, CHEAR reference urine, NIST SP:SAMPLEPREP_SUMMARY reference urine, and blanks were mixed with 400 µL methanol containing 500 SP:SAMPLEPREP_SUMMARY ng/mL tryptophan-d5 as internal standard and vortexed by a multiple tube vortex SP:SAMPLEPREP_SUMMARY mixer for 2 min at 5000 rpm at room temperature. All samples were centrifuged at SP:SAMPLEPREP_SUMMARY 16,000 rcf for 10 min at 4°C. The supernatant (350 µL) was transferred into a SP:SAMPLEPREP_SUMMARY pre-labeled 2.0 mL Lo-bind Eppendorf tube, dried by a SpeedVac overnight, and SP:SAMPLEPREP_SUMMARY stored at -80° C. For immediate analysis, 100 µL of water-methanol solution SP:SAMPLEPREP_SUMMARY (95:5, v/v) was used to reconstitute the dried extracts.
