Here, we present a dual stage controlled clinical intervention study PR:PROJECT_SUMMARY with healthy individuals challenged with lipopolysaccharide. While in a first PR:PROJECT_SUMMARY stage, plasma proteomics and lipidomics was applied to observe the kinetics of PR:PROJECT_SUMMARY inflammatory modulators within eight hours, the effects of a placebocontrolled PR:PROJECT_SUMMARY anti-oxidative intervention were determined in the second stage. Plasma proteome PR:PROJECT_SUMMARY profiling demonstrated the early involvement of platelets detectable within two PR:PROJECT_SUMMARY hours after lipopolysaccharide challenge, followed by the characteristic PR:PROJECT_SUMMARY induction of liver-derived acute phase proteins and innate immune cell-derived PR:PROJECT_SUMMARY alarmins. Untargeted lipidomics demonstrated the early release of fatty acids PR:PROJECT_SUMMARY and taurocholic acid within two hours, followed by complex time courses of PR:PROJECT_SUMMARY various oxylipins and the downregulation of numerous lysophospholipids and PR:PROJECT_SUMMARY deoxycholic acid. Groups of molecules with similar kinetics during the time PR:PROJECT_SUMMARY course analysis upon lipopolysaccharide challenge were observed to have common PR:PROJECT_SUMMARY precursors or synthesizing enzymes. Dietary supplementation with antioxidants PR:PROJECT_SUMMARY did not affect the kinetics of detectable proteins, but significantly PR:PROJECT_SUMMARY downregulated the pro-inflammatory sphingosine-1-phosphate and increased the PR:PROJECT_SUMMARY levels of oxylipins described to facilitate the resolution of inflammation, PR:PROJECT_SUMMARY 20-HEPE and 22-HDoHE. For TR:TREATMENT_SUMMARY this randomized, double-masked, placebo-controlled parallel group study, 30 TR:TREATMENT_SUMMARY healthy male human individuals were included upon passing a screening TR:TREATMENT_SUMMARY examination and signed written informed consent prior to study entry. Subjects TR:TREATMENT_SUMMARY with any clinically relevant illness, intake of medication, including vitamin or TR:TREATMENT_SUMMARY mineral supplements, or blood donation within 3 weeks prior to the study were TR:TREATMENT_SUMMARY excluded and subjects who did not complete the study were replaced. In addition, TR:TREATMENT_SUMMARY participants had to abstain from alcohol or caffeine containing beverages within TR:TREATMENT_SUMMARY 12 hours prior to each study day of the dual stage controlled clinical TR:TREATMENT_SUMMARY intervention study. To induce systemic inflammation and oxidative stress, an TR:TREATMENT_SUMMARY intravenous infusion of a bolus containing 2 ng/kg bodyweight Escherichia coli TR:TREATMENT_SUMMARY endotoxin known as lipopolysaccharide (LPS; NIH-CC, Bethesda, MD, USA) was used. After this first stage, participants TR:TREATMENT_SUMMARY were randomly assigned to take either the omega-3 fatty acid containing food TR:TREATMENT_SUMMARY supplement Vitamac (n = 17; Croma Pharma GmbH, Korneuburg, Austria) or matching TR:TREATMENT_SUMMARY lactose and wheat starch containing placebo capsules (n = 13) for 14 days. After TR:TREATMENT_SUMMARY the 14 days intervention, all participants were treated again with LPS in the TR:TREATMENT_SUMMARY second stage and plasma was collected at baseline (BL), 60 min, 120 min, 240 min TR:TREATMENT_SUMMARY and 480 min after LPS infusion applying the same protocol as described for the TR:TREATMENT_SUMMARY first stage. #SAMPLEPREP SP:SAMPLEPREP_SUMMARY Frozen EDTA-anticoagulated plasma was freshly thawed on ice. The MS scan range was 250-700 m/z with a resolution of 60,000 (at m/z MS:MS_COMMENTS 200) on the MS1 level. A Top 2 method was applied for fragmentation (HCD 24 MS:MS_COMMENTS normalized collision energy). The resulting fragments were analysed on the MS2 MS:MS_COMMENTS level at a resolution of 15,000 (at m/z 200). Operating in positive ionization MS:MS_COMMENTS mode, a spray voltage of 3.5 kV and a capillary temperature of 253°C were MS:MS_COMMENTS applied.
