Despite PR:PROJECT_SUMMARY significant efforts to control and eliminate the disease, malaria remains a PR:PROJECT_SUMMARY leading cause of illness and death, mainly due to the occurrence of PR:PROJECT_SUMMARY drug-resistant parasites to the frontline antimalarials such as PR:PROJECT_SUMMARY dihydroartemisinin-piperaquine (DHA-PPQ). Partial artemisinin resistance has PR:PROJECT_SUMMARY been linked to kelch13 mutations, while decreased PPQ sensitivity has been PR:PROJECT_SUMMARY associated with higher plasmepsin II and III gene copies and mutations in the PR:PROJECT_SUMMARY chloroquine resistance transporter. In this study, we demonstrate the effective PR:PROJECT_SUMMARY use of CRISPR/Cas9 technology to generate single knockouts (KO) of plasmepsin II PR:PROJECT_SUMMARY and plasmepsin III, as well as a double KOs of both genes, in two isogenic lines PR:PROJECT_SUMMARY of Cambodian parasites with varying numbers of plasmepsin gene copies. The PR:PROJECT_SUMMARY deletion of plasmepsin II and/or III increased the parasites' sensitivity to PR:PROJECT_SUMMARY PPQ, evaluated by the area under the curve. We explored several hypotheses to PR:PROJECT_SUMMARY understand how an increased plasmepsin gene copy number might influence parasite PR:PROJECT_SUMMARY survival under high PPQ pressure. Our findings indicate that protease inhibitors PR:PROJECT_SUMMARY have a minimal impact on parasite susceptibility to PPQ. Additionally, parasites PR:PROJECT_SUMMARY with higher plasmepsin gene copy numbers did not exhibit significantly increased PR:PROJECT_SUMMARY hemoglobin digestion, nor did they produce different amounts of free heme PR:PROJECT_SUMMARY following PPQ treatment compared to wildtype parasites. Interestingly, PR:PROJECT_SUMMARY hemoglobin digestion was slowed in parasites with plasmepsin II deletions.
