Inflammatory bowel diseases (IBD) patients have PR:PROJECT_SUMMARY altered gut microbial metabolic signatures driven by shifts in the microbiome. PR:PROJECT_SUMMARY However, the microbial metabolites driving inflammation and remission and their PR:PROJECT_SUMMARY immunomodulatory roles remain largely uncharacterized. We prioritized metabolic PR:PROJECT_SUMMARY features from untargeted IBD fecal metabolomics and selected candidate features PR:PROJECT_SUMMARY based on their microbial association for structural elucidation. We uncovered PR:PROJECT_SUMMARY four acylated putrescines that are IBD-associated and microbially-dependent and PR:PROJECT_SUMMARY investigated their bioactivity. One acylated putrescine, N-oleoylputrescine PR:PROJECT_SUMMARY (NOP), has anti-inflammatory activity and suppressed type 1 immune responses. PR:PROJECT_SUMMARY NOP ameliorated inflammation in several colitis models, modulating myeloid and PR:PROJECT_SUMMARY lymphocyte cell populations and dampening proinflammatory responses, relevant PR:PROJECT_SUMMARY for resolution of inflammation and maintenance of remission in IBD. This mouse fecal microbiome data was used to develop a filter ST:STUDY_SUMMARY and prioritization tool: Metabolome Analysis and Combined Annotation Ranks to ST:STUDY_SUMMARY pRioritize Novel bioactives, that we call micro-MACARRoN.
