Internal Standard: Twenty-one internal standards were obtained from SP:SAMPLEPREP_SUMMARY Cambrige Isotope Labs (Andover, Massachusettes, USA). Individual internal SP:SAMPLEPREP_SUMMARY standard stock solutions were prepared by dissolving 1-2 mg (accurately weighed) SP:SAMPLEPREP_SUMMARY dry standard in appropriate solvents (Appendix 1). The final internal standard SP:SAMPLEPREP_SUMMARY solution was generated by mixing stock solutions and diluting with methanol. The SP:SAMPLEPREP_SUMMARY internal standards solutions were prepared as described in Appendix 1 (all SP:SAMPLEPREP_SUMMARY compounds with ID starting with ‘is’). The solutions were stored at -80°C. SP:SAMPLEPREP_SUMMARY Preparation Blank: Methanol (240 uL) used in the above sample prep was mixed SP:SAMPLEPREP_SUMMARY with 60 µL of IS. Solvent Blank: The following solvents that were used in the SP:SAMPLEPREP_SUMMARY above sample prep, 300 µL of MMA, 300 µL of water, 300 µL of methanol, were SP:SAMPLEPREP_SUMMARY mixed without the addition of IS. Generation of Benchmark Samples: All samples SP:SAMPLEPREP_SUMMARY were prepared by mixing plasma extracts, yeast extracts, spikeA, spikeB, and IS SP:SAMPLEPREP_SUMMARY according to the volume ratios presented in Appendix 3 into respective glass SP:SAMPLEPREP_SUMMARY autosampler vials (2 mL). When the volume of assigned solutions was greater than SP:SAMPLEPREP_SUMMARY 1.8 mL, reagents were added sequentially with intermittent removal of solvents SP:SAMPLEPREP_SUMMARY under nitrogen. The full volume of added reagent described in Appendix 3 was SP:SAMPLEPREP_SUMMARY dried in the vial, and resuspending in 1500 µL of Methanol and vortex mixed to SP:SAMPLEPREP_SUMMARY resuspend solutes. Of these 1500 uL, 600 uL were transferred to a Washington SP:SAMPLEPREP_SUMMARY University vial, and 800 uL were transferred to a new CSU vial. Solvent was SP:SAMPLEPREP_SUMMARY again evaporated under nitrogen in preparation for shipping, and stored in SP:SAMPLEPREP_SUMMARY -80°C until shipment. Samples were reconstituted in absolute methanol prior to SP:SAMPLEPREP_SUMMARY analysis, at equal concentration to the original aliquot volume prior to drying. SP:SAMPLEPREP_SUMMARY For GC-MS, solvent was evaporated under nitrogen. The dried samples were treated SP:SAMPLEPREP_SUMMARY with 30 uL of methoxyamine hydrochloride in pyridine (25 mg/mL), vortexed and SP:SAMPLEPREP_SUMMARY heated at 60°C for 45 min. The vials were then sonicated for 10 min and SP:SAMPLEPREP_SUMMARY incubated again for another 45 min.
