The carbon-13 labeled treatments were at TR:TREATMENT_SUMMARY equivalent concentrations to complete RPMI1640. For each treatment type (glucose TR:TREATMENT_SUMMARY or glutamine), every sample was treated at the same time and collected at either TR:TREATMENT_SUMMARY 0.25 (15 minutes), 6, 16, or 24 hours and were flash frozen in liquid N2 until TR:TREATMENT_SUMMARY metabolite extraction. For the inhibitor experiments, cells were pre-incubated TR:TREATMENT_SUMMARY with inhibitors for 20 minutes in either glucose-free or glutamine-free RPMI TR:TREATMENT_SUMMARY before RPMI1640 equivalent concentration of either U-13C-glucose or TR:TREATMENT_SUMMARY U-13C-glutamine was spiked into each sample. The cells were then co-cultured TR:TREATMENT_SUMMARY with the inhibitors and either U-13C-glucose or -glutamine for 6 hours. After TR:TREATMENT_SUMMARY the co-culture, cells were collected and flash frozen in liquid N2 until TR:TREATMENT_SUMMARY metabolite extraction. #SAMPLEPREP SP:SAMPLEPREP_SUMMARY Frozen pellets were removed from storage and placed on wet ice. Metabolites were SP:SAMPLEPREP_SUMMARY then extracted from samples by adding 1mL of -80°C 80:20 MeOH:H2O to each tube, SP:SAMPLEPREP_SUMMARY vigorously vortexing, and incubating samples in -80°C freezer for >15 minutes. SP:SAMPLEPREP_SUMMARY Once metabolites were extracted, precipitated protein was pelleted via SP:SAMPLEPREP_SUMMARY centrifugation at max speed for 10 minutes at 4°C. Extracted metabolites in the SP:SAMPLEPREP_SUMMARY supernatant were removed and placed in a new tube and dried under N2. Pelleted SP:SAMPLEPREP_SUMMARY proteins were resolubilized to determine protein concentration via a BCA assay. SP:SAMPLEPREP_SUMMARY Once samples were dry, metabolites were resolubilized in 50uL of 3:2 mobile SP:SAMPLEPREP_SUMMARY phase A:mobile phase B (mobile phase A: 20mM NH4OAc, 20mM NH4OH, 5% acetonitrile SP:SAMPLEPREP_SUMMARY in H2O; mobile phase B: 100% acetonitrile). Once resuspended, samples were spun SP:SAMPLEPREP_SUMMARY down at max speed for 10 minutes at 4°C, transferred to a new tube and spun SP:SAMPLEPREP_SUMMARY again to remove all insoluble debris. After the second spin, samples were added SP:SAMPLEPREP_SUMMARY to vial inserts and were ready to be run via LCMS/MS.
