In this study, we PR:PROJECT_SUMMARY investigate the temporal and spatial dynamics of phytoplankton community and the PR:PROJECT_SUMMARY production of their primary and secondary untargeted metabolites in response to PR:PROJECT_SUMMARY local meteorological events. Thus, we collected water samples in two points of PR:PROJECT_SUMMARY the Aydat Lake (France): near the inflowing waters from Veyre River and at the PR:PROJECT_SUMMARY middle of the lake during the 2021 summer. Untargeted intracellular metabolites PR:PROJECT_SUMMARY were measured using ultra-high-performance liquid chromatography coupled with a PR:PROJECT_SUMMARY high-resolution mass spectrometer, as well as phytoplankton biovolume and PR:PROJECT_SUMMARY diversity and physicochemical lake’s parameters. Primarily, our results show PR:PROJECT_SUMMARY the increase of the biovolume of diazotrophic cyanobacteria at the end of the PR:PROJECT_SUMMARY drought and after rain events at both sites. During the drought, we observe a PR:PROJECT_SUMMARY strong increase of intracellular lipid contents, probably in response to sudden PR:PROJECT_SUMMARY nitrogen and phosphorus limitation. Differently, during the wet periods, we PR:PROJECT_SUMMARY observe an increase of the phytoplankton glycerophospholipid content, especially PR:PROJECT_SUMMARY at the middle of the lake, whereas significantly higher abundance of secondary PR:PROJECT_SUMMARY metabolites was monitored at site near the wetland area. Some of these PR:PROJECT_SUMMARY metabolites correspond to Cyanopepetids, strongly correlated with the biovolume PR:PROJECT_SUMMARY of Dolichospermum, present at both sites. Therefore, these results suggest PR:PROJECT_SUMMARY adaptative responses to cope with the phytoplankton growing stimulation related PR:PROJECT_SUMMARY with the increase of the nutritive ion influx following the rain events. At each depth and location, approximately 10 liters of CO:COLLECTION_SUMMARY lake water collected with a Van Dorn bottle are then pre-filtered through a CO:COLLECTION_SUMMARY 250-micrometer Nylon filter to remove larger particles and kept phytoplankton, CO:COLLECTION_SUMMARY and then transferred into 15-liter containers.
