The CO:COLLECTION_SUMMARY supernatant (plasma) was carefully aliquoted and immediately frozen at -80°C. CO:COLLECTION_SUMMARY All procedures were performed with samples wrapped in aluminum foil to prevent CO:COLLECTION_SUMMARY light exposure. CO:SAMPLE_TYPE Blood (plasma) #TREATMENT TR:TREATMENT_SUMMARY The plasma collected for subsequent detection primarily includes samples from TR:TREATMENT_SUMMARY the following treatment groups: (1) SHAM group: Plasma derived from mice that TR:TREATMENT_SUMMARY underwent sham surgery without any intervention; (2) AAC group: Plasma derived TR:TREATMENT_SUMMARY from mice subjected to abdominal aortic constriction (AAC) to induce a TR:TREATMENT_SUMMARY pressure-overload myocardial hypertrophy model;(3)AAC+Ex group: Plasma TR:TREATMENT_SUMMARY derived from mice that underwent AAC surgery and were then subjected to TR:TREATMENT_SUMMARY moderate-intensity aerobic exercise; (4) AAC+TPPU group: Plasma derived from TR:TREATMENT_SUMMARY mice that underwent AAC surgery and were administered 10 mg/L TR:TREATMENT_SUMMARY 1-trifluoromethoxyphenyl-3-(1-propionylpiperidin-4-yl) urea (TPPU) in drinking TR:TREATMENT_SUMMARY water; (5) AAC+Ex+AAV-sEH group: Plasma derived from mice that underwent AAC TR:TREATMENT_SUMMARY surgery, were subjected to moderate-intensity aerobic exercise, and concurrently TR:TREATMENT_SUMMARY received an intraperitoneal injection of AAV-soluble epoxide hydrolase (sEH). #SAMPLEPREP SP:SAMPLEPREP_SUMMARY Sample preparation was performed using a solid-phase extraction unit connected SP:SAMPLEPREP_SUMMARY to an HLB column, and plasma samples pre-spiked with internal standards were SP:SAMPLEPREP_SUMMARY adsorbed over the column using 5% methanol after activation with ethyl acetate SP:SAMPLEPREP_SUMMARY and methanol, and subsequently extracted using methanol and ethyl acetate after SP:SAMPLEPREP_SUMMARY vacuum drying. The obtained samples were dried under nitrogen and re-solubilized SP:SAMPLEPREP_SUMMARY using 30% acetonitrile on the machine. SP:PROCESSING_METHOD solid-phase extraction SP:PROCESSING_STORAGE_CONDITIONS -80℃ SP:EXTRACTION_METHOD quantitative 200ul of plasma were taken, and 1 μl of the mixed eico-IS was SP:EXTRACTION_METHOD added to each sample, followed by connecting a Waters HLB 3 cc (60 mg) SP:EXTRACTION_METHOD extraction cartridge to a solid-phase extraction device (Supelco, USA). The SP:EXTRACTION_METHOD extraction procedure involved passing 2 ml of ethyl acetate through the SP:EXTRACTION_METHOD cartridge, then passing 2 ml of methanol through the cartridge, followed by 2 ml SP:EXTRACTION_METHOD of 5% methanol (containing 0.1% acetic acid), and 2 ml of supernatant, and again SP:EXTRACTION_METHOD passing 2 ml of 5% methanol (containing 0.1% acetic acid) through the cartridge. SP:EXTRACTION_METHOD The samples were then dried under vacuum for 15 minutes. A new 2 ml EP tube was SP:EXTRACTION_METHOD prepared for sample collection, 0.6 ml of methanol and 1.4 ml of ethyl acetate SP:EXTRACTION_METHOD were added through the cartridge. The organic phase was then evaporated to SP:EXTRACTION_METHOD dryness under a stream of high purity nitrogen. Following this, 100 μl of 30% SP:EXTRACTION_METHOD acetonitrile was added and vortexed 10 min for re-dissolution. After vigorous SP:EXTRACTION_METHOD mixing, samples were filtered by use of centrifuge tube filters (nylon membrane, SP:EXTRACTION_METHOD 0.22 μm).
