admitted to the Department of CO:COLLECTION_SUMMARY Obstetrics and Gynecology at Inönü University Turgut Özal Medical Center and CO:COLLECTION_SUMMARY healthy pregnant women without preterm labor as controls. For each participant, CO:COLLECTION_SUMMARY samples were collected in two separate tubes for metagenomics and metabolomics CO:COLLECTION_SUMMARY analyses and stored at –80°C until processing. Ethical approval was obtained CO:COLLECTION_SUMMARY from the Malatya Clinical Research Ethics Committee (2020/91), and all CO:COLLECTION_SUMMARY participants provided written informed consent. PPROM diagnosis was confirmed by CO:COLLECTION_SUMMARY amniotic fluid leakage observed during a sterile speculum exam or a positive CO:COLLECTION_SUMMARY AmniSure test. Exclusion criteria included systemic diseases (e.g., CO:COLLECTION_SUMMARY hypertension, diabetes), pregnancy complications (e.g., gestational diabetes, CO:COLLECTION_SUMMARY hypertensive disorders), fetal anomalies, prior urogenital surgery, CO:COLLECTION_SUMMARY urolithiasis, congenital anomalies, signs of acute fetal distress, or recent CO:COLLECTION_SUMMARY antibiotic/probiotic use within one month. Control participants were healthy CO:COLLECTION_SUMMARY pregnant women without systemic disease, preterm labor, vaginal bleeding, or CO:COLLECTION_SUMMARY recent antibiotic/probiotic exposure. To prevent contamination, urine was CO:COLLECTION_SUMMARY collected via sterile catheterization and immediately stored at –80°C. CO:SAMPLE_TYPE Urine #TREATMENT TR:TREATMENT_SUMMARY Urine samples stored at –80°C were thawed at room temperature before ¹H NMR TR:TREATMENT_SUMMARY analysis. Each 400 µL sample was mixed with 200 µL of phosphate buffer (0.2 M TR:TREATMENT_SUMMARY Na₂HPO₄ and 0.2 M KH₂PO₄ in D₂O, pH 7.4) for pH stabilization and TR:TREATMENT_SUMMARY field-frequency locking. The mixture was centrifuged at 5000 × g for 5 minutes TR:TREATMENT_SUMMARY at 5°C, and 550 µL of the supernatant was transferred into 5 mm NMR tubes. #SAMPLEPREP SP:SAMPLEPREP_SUMMARY Analyses were performed using a Bruker Avance III HD 600 MHz spectrometer SP:SAMPLEPREP_SUMMARY (Bruker GmBH, Germany) with a 5 mm broadband BBO probehead (¹H/¹³C/¹⁵N) at SP:SAMPLEPREP_SUMMARY the Inönü University Scientific and Technological Research Center. Sample and SP:SAMPLEPREP_SUMMARY probe temperatures were stabilized at 296 K with a Bruker Cooling Unit (BCU). SP:SAMPLEPREP_SUMMARY The ¹D-NOESY-presaturation (noesygppr1d, Bruker) pulse sequence (RD ‒ 180° SP:SAMPLEPREP_SUMMARY ‒ {mixing} ‒ 90° ‒ FID) was applied to suppress water and protein-derived SP:SAMPLEPREP_SUMMARY signals. The spectra were recorded with a spectral width of 7002.8 Hz, 32 K data SP:SAMPLEPREP_SUMMARY points, and a 40 ms mixing time. Additionally, weak presaturation pulses SP:SAMPLEPREP_SUMMARY (1.1E-4W power) were applied to enhance water signal suppression.
