Many of these diseases are influenced by PR:PROJECT_SUMMARY age-associated metabolic dysregulation, but how metabolism changes in multiple PR:PROJECT_SUMMARY organs during aging in males and females is not known. Answering this could PR:PROJECT_SUMMARY reveal new mechanisms of aging and age-targeted therapeutics. In this study, we PR:PROJECT_SUMMARY describe how metabolism changes in 12 organs in male and female mice at 5 PR:PROJECT_SUMMARY different ages. Organs show distinct patterns of metabolic aging that are PR:PROJECT_SUMMARY affected by sex differently. Hydroxyproline shows the most consistent change PR:PROJECT_SUMMARY across the dataset, decreasing with age in 11 out of 12 organs investigated. We PR:PROJECT_SUMMARY also developed a metabolic aging clock that predicts biological age and PR:PROJECT_SUMMARY identified alpha-ketoglutarate, previously shown to extend lifespan in mice, as PR:PROJECT_SUMMARY a key predictor of age. CO:COLLECTION_SUMMARY The pancreata were either not eligible for transplant or they had no eligible CO:COLLECTION_SUMMARY recipients. While maintaining blood flow, we cross-clamped the superior CO:COLLECTION_SUMMARY mesenteric and celiac arteries, recovered the pancreas and immediately placed it CO:COLLECTION_SUMMARY in a physiologic organ preservation solution on ice. We then transported the CO:COLLECTION_SUMMARY pancreata to the University of Michigan, approximately 6 miles away, where it CO:COLLECTION_SUMMARY was dissected into head, body and tail, and then immediately flash frozen in CO:COLLECTION_SUMMARY liquid nitrogen. TR:TREATMENT_SUMMARY Confounding factors, such as donor diseases (e.g., diabetes and obesity), made TR:TREATMENT_SUMMARY these organs ineligible for transplantation, which rendered them available for TR:TREATMENT_SUMMARY research. #SAMPLEPREP SP:SAMPLEPREP_SUMMARY For tissue homogenization, we put tissue samples in 10 μL ice-cold 4:1 LC-grade SP:SAMPLEPREP_SUMMARY Methanol:Ultrapure-water per mg tissue and homogenized them using a bead SP:SAMPLEPREP_SUMMARY homogenizer. We centrifuged tissue homogenates at 4°C at 21,300 g for 10 SP:SAMPLEPREP_SUMMARY minutes and collected the metabolite-containing supernatants.
