The oxPPP generates NADPH at two PR:PROJECT_SUMMARY steps, glucose-6-phosphate dehydrogenase (G6PD), the most common enzyme PR:PROJECT_SUMMARY deficiency in humans, and 6-phosphogluconate dehydrogenase (PGD). Previous PR:PROJECT_SUMMARY literature suggests that G6PD supports but PGD limits T cell-mediated immunity. PR:PROJECT_SUMMARY Here we use T cell-specific knockout mouse models to show that both enzymes are PR:PROJECT_SUMMARY required for anti-tumor immunity and response to immunotherapy. PGD knockout PR:PROJECT_SUMMARY depletes mature T cells systemically, while G6PD loss does not reduce basal T PR:PROJECT_SUMMARY cell populations but results in apoptosis upon activation. Such apoptosis is not PR:PROJECT_SUMMARY reversed by major downstream products of the oxPPP, including antioxidants, PR:PROJECT_SUMMARY nucleosides, or fatty acids. Instead, T cells are partially rescued by removal PR:PROJECT_SUMMARY of media cystine, whose reduction requires NADPH. G6PD loss induces an oxidative PR:PROJECT_SUMMARY stress response that upregulates cystine import, which together with low NADPH PR:PROJECT_SUMMARY leads to fatal disulfide stress. Naïve CO:COLLECTION_SUMMARY T cells were cultured at 1 × 10^6 cells ml-1 in complete RPMI medium (RPMI-1640 CO:COLLECTION_SUMMARY medium, 10% heat inactivated complete FBS (35-011-CV, Corning), 100 U ml−1 CO:COLLECTION_SUMMARY penicillin, 100 µg ml−1 streptomycin (15140-122, Gibco) and 55 μM CO:COLLECTION_SUMMARY 2-mercaptoethanol (21985-023, Gibco)) and activated with plate-bound anti-CD3 CO:COLLECTION_SUMMARY (10 µg ml−1; BE0001-1, Bio X Cell), anti-CD28 (5 µg ml−1; CO:COLLECTION_SUMMARY BE0015-1, Bio X Cell) and soluble recombinant murine IL-2 (20 ng ml−1; CO:COLLECTION_SUMMARY 212-12, Peprotech) for 24 h. While irrelevant for metabolomics, for the last 2 CO:COLLECTION_SUMMARY h, tracing medium containing 100% [1,2-13C] glucose (CLM-504, Cambridge Isotope CO:COLLECTION_SUMMARY Laboratories, Inc.) was spiked in to achieve 20% glucose labeling.
