Collection Summary:Healthy adolescent, male Yorkshire swine (average weight 50.0 ± 2.7 kg) were acclimated in the animal facilities for a minimum of 3 days and examined by a veterinarian on the day of experimentation. In brief, on the day of surgery, swine anesthesia was induced via intramuscular injection of ketamine (20.0 mg/kg), xylazine (2.0 mg/kg), and acepromazine (0.2 mg/kg) followed by intubation under direct laryngoscopy. Continuous infusion anesthesia was maintained through a superficial ear vein using propofol (3 mg/kg/h) and fentanyl (3 mcg/kg/h). Lactated Ringer’s solution was administered at a rate of 10 mL/kg/h to overcome insensible losses. Mahurkar catheters (12 Fr) were placed in the femoral arteries and veins for vascular access.

Treatment Summary:the DCBI model included blast traumatic brain injury (bTBI), tissue injury through bilateral open femur fractures, followed by severe fixed pressure targeted hemorrhagic shock (HS). Zone-I aortic occlusion (AO) was performed using the ER-REBOA Catheter (Prytime Medical Devices, Inc., Boerne, TX). The catheter was inserted through a 7 Fr sheath placed in the common femoral artery, was advanced to 50 cm, and inflated using 6 mL of normal saline for Zone-I AO and placed at 26 cm with 5 ml of saline for Zone-III, and both were maintained for the final 30 minutes of the HS phase.

Sampleprep Summary:Plasma samples were thawed on ice then 20 μl was aliquoted into individual 1.5mL microcentrifuge tubes. The plasma was diluted to 480 μl with methanol:acetonitrile:water (5:3:2, v/v/v). The metabolite containing supernatant was transferred to individual auto sampler vials on ice.
