Study SummaryThe skin microbiota aid in the skin’s barrier function and help mature our immune system as well as produce odor by metabolizing sweat and other secretions produced by our bodies. Only now with the advancement of high throughput sequencing technologies has it become possible to sample the diversity of the microbiota that inhabits the outer surface of the body. This project will sample DNA from microbes (single celled organisms) that live on the human skin, investigate microbial gene regulation, skin surface metabolites, microbial odor production, and begin to assess the human perception of pleasantness of these microbial associated odors.

Project Summary:Diverse communities of microscopic organisms (microbiota) live on the skin, and their collective genomes are referred to as the skin microbiome. The skin microbiota aid in the skin’s barrier function and help mature our immune system as well as produce odor by metabolizing sweat and other secretions produced by our bodies. Sampleprep Summary:Extracted sweat from cotton balls were mixed with 2mL 3:3:2 ACN:IPA:H2O by vortex mixer then centrifuged. Supernatant recovered. Archived 450µL of supernatant. Continued prep of remaining 450µL supernatant. Evaporated to dryness w/speed vac. Formation of methoximes by adding 10µL of 40mg/mL MeOx in pyridine to each sample. Placed onto Thermomixer for 90 min at 30⁰C. Then FAME retention index markers and MSTFA added to each sample for derivatization step. Samples returned to Thermomixer for 45 min at 70⁰C.
