Sampleprep Summary:Metabolite Extraction. Mouse small intestine digesta samples (luminal contents) were collected, weighed and homogenized in water followed by addition of acetonitrile for metabolite extraction. After vortex mixing, 30 second sonication, and centrifugation, the clear supernatants were collected and dried under a gentle nitrogen gas flow. The dried metabolite residues were dissolved in 40% acetonitrile containing 0.01% formic acid, 4 ?L per mg of the raw material. After 15 sec vortex and 30 sec sonication, each sample was further diluted 1:2 with water. Twelve-?L aliquots were injected for untargeted metabolic fingerprinting by ultrahigh-performance liquid chromatography ? Fourier transform mass spectrometry (UPLC-FTMS).
