Sampleprep Summary:In the allocated randomized order, frozen plasma samples were thawed on ice and analyzed in five analytical batches. Each batch consisted of a set of QA/QC samples which included the HHEAR untargeted plasma pools 1 and 2, NIST standard reference material (NIST 1958), and procedural blanks. Briefly, all samples were prepared for analysis in 5 mL glass tubes by adding 20 μL of MS grade formic acid to 100μL of plasma (or QA/QC sample or procedural blanks). Following this, a 4:1 Hexane:Ethyl acetate solution spiked with 10 ng mL-1 of internal standard was added as the extraction solvent and the samples vortexed. The samples were then centrifuged and the supernatant transferred to a new glass tube containing about 25 mg of dispersive SPE (dSPE) powder for water removal and extraction cleanup. The tubes were then vigorously agitated for 5 min, centrifuged, and the supernatant transferred to vials for GC-HRMS analysis.
