Study SummaryMCJ/DnaJC15 is an endogenous negative regulator of Complex I and mitochondrial respiration. The goal of these experiments are to characterize the metabolic profile of murine WT or MCJ KO CD8 CD19-BBz CAR-T cells in cytokine-deprived condition. The CAR-T cells were expanded with IL-2 (60IU/ml) for 3 expansions (6 days) and rested in the medium for 48 hours.

Collection Summary:Murine CD8 cells were isolated from lymph nodes and spleen using negative selection with CD8+ T cell isolation kit (for generation of CAR-T cells) (Miltenyi, order# 130-104-075) or positive selection (for OT-I CD8 cells) with CD8 (Ly-2) Microbeads (Miltenyi, order# 130-117-044). Mouse CD8 cells were activated with plate-bound anti-CD3 (5 g/ml) and soluble anti-CD28 mAbs (1 g/ml) for 48h for WT and cMCJ KO CD8 studies and OT-I CD8 cells studies.
